Wales have not given up hope of fly-half Sam Costelow still playing a part in the Six Nations but flanker Taine Plumtree has been ruled out of the tournament.
Both players were injured during the 26-23 defeat by Scotland on Saturday.
Head coach Steve Tandy admitted afterwards they did not look good with the Scarlets duo having scans on Monday.
Costelow, 25, left the field in the second half with an ankle problem but the Scarlets playmaker is staying with the squad to “continue with the rehabilitation” ahead of games with Ireland and Italy.
Back rower Plumtree, also 25, was forced off with a shoulder problem after just nine minutes and has been released by Wales.
Ospreys lock Rhys Davies was ruled out earlier this week after suffering an ankle injury in training.
No players have been called up yet to cover for the injured trio with the official squad number consisting of 37 players.
There could be some additions when they all come back together on Sunday.
Wales have released nine players back to their regions this weekend to play in United Rugby Championship (URC) matches, while the English and French-based players are meant to return to their clubs.
Wales are bottom of the Six Nations table after losses to England, France and Scotland with their three-year losing sequence in the tournament stretching to 14 games.
Tandy’s side travel to face Ireland in Dublin on Friday, 6 March before hosting Italy in the tournament finale eight days later.

There is still a major doubt whether Costelow will play any further role in the tournament.
After being recalled to face Scotland, the fly-half impressed in the 55 minutes that he was on the field at the Principality Stadium.
“He’s been through a lot in his Wales career with not being involved in the autumn but we backed him to come back in and he really took that opportunity,” stated Tandy.
“He was fantastic with the way he drove the team forward in that first half. It’s such a shame for him that it ended with injury.”
Costelow had replaced Ospreys fly-half Dan Edwards, who had started the previous seven Wales internationals, including six under Tandy.
Edwards, 22, was dropped from the match-day 23 against Scotland with Harlequins fly-half Jarrod Evans coming on as a replacement.
Centres Joe Hawkins and Ben Thomas have also had experience of playing 10.

Wales have lost two back five forwards this week so will be looking at their second row and back row resources.
Cardiff flanker James Botham impressed when he came off the replacements bench for Plumtree as he formed the back-row trio with Alex Mann and Aaron Wainwright, who has been Wales’ player of the tournament so far.
Other options in the squad include Olly Cracknell and Josh Macleod, who have both started a game in the Six Nations, and Ospreys open-side flanker Harri Deaves, who was used as a replacement against England.
Leicester flanker Tommy Reffell has been ignored so far by Tandy after being left out of two squads.
Other potential call-ups include Ospreys duo Morgan Morse and Ross Moriarty and Dragons flanker Ryan Woodman, who can also double up at lock.
Dafydd Jenkins and Ben Carter have established themselves as Wales’ first-choice second-row pairing after their dynamic displays against Scotland with 22 tackles each.
They are backed up by Adam Beard, who was dropped after the 54-12 loss to France and Gloucester’s Freddie Thomas, who was a replacement last weekend.
